titleOfInvention | Method of screening substance with anticancerous activity and apparatus therefor |
abstract | In one preferred embodiment, a sensor chip having cells cultured therein is set in a measuring part. The measuring part is filled with a culture medium, and measuring is initiated. Culture medium replacement is carried out 10 min later with culture mediums respectively containing 10, 25, 50 and 100 μM quercetin, resveratrol and rutin and, as control, a culture medium containing DMSO, and measuring is continued. After the initiation of measuring, the from 2700 to 3000 sec 5 min time-rate-of-change of surface plasmon resonance angle is determined, and the correlation thereof with cell survival rate is determined. Thus, there has been obtained a linear relationship. Using this result as a working curve, the anticancerous activity can be quantitatively assessed by measuring the specified brief time-rate-of-change of surface plasmon resonance angle. Actually, when using extracted component luteolin as other substance with anticancerous activity and herceptin as an antibody anticancer agent, the time-rate-of-change has been worked out from the survival rate and surface plasmon resonance angle measurements, the result agreeing with the above working curve has been obtained. |
