abstract A method capable of detecting a target antigen with high sensitivity. In particular, a method of detecting a target antigen, comprising the step (a) of reacting a target antigen with one or two or more antibodies to thereby form an antigen antibody composite containing the target antigen and the antibodies; the step (b) of before, during or after the step (a), binding a single strand nucleic acid of base sequence represented by any one of the general formulae (I) to (IV) at the 5’ end to the antibodies; the step (c) of after the steps (a) and (b), causing a DNA polymerase to act on the single strand nucleic acid in the presence of deoxynucleoside triphosphates containing deoxyadenosine triphosphate and deoxythymidine triphosphate and/or deoxyuridine triphosphate at such a temperature that formation of the antigen antibody composite can be maintained; and the step (d) of after the step (c), detecting self-extension of the single strand nucleic acid bound to the antibodies as a constituent of the antigen antibody composite.
