abstract | There is provided for the separation of one or more protein(s) contained in host cells comprising isolating the protein(s) and solubilising, unfolding and refolding the isolated protein(s) wherein an effective amount of a non-ionic surfactant is introduced during the solubilising, unfolding or refolding. Preferably, the protein(s) which may be separated from host cells by the process of the present invention include bacterial and plant derived toxin protein(s), or analogues thereof. Non-ionic surfactants that may be employed in the process include alkyl and alkaryl capped polyoxyalkylenes, particularly alkyl and alkaryl capped polyoxyethylenes, mixed polyoxyalkylene, particularly polyoxyethylene-polyoxypropylene-polyoxyethylenes, and polyoxyalkylene sorbitan carboxylates, particularly polyoxyethylene sorbitan carboxylates. Advantageously, the protein(s) and compositions comprising protein(s) and non-ionic surfactant obtainable by these processes show improvement in the ability to stimulate an immune response. |
