abstract The invention relates to a method for the isolation of plasmid-DNA from biomasses, by means of an aqueous biphasic system, comprising a polymer and a salt component, characterised in that the resuspension of the applied biomass, the alkaline lysates of the biomass, the neutralisation of the alkaline lysate component and the separation of the plasmid-DNA from the contaminants (such as cell remains, RNA and gDNA, for example) is carried out in a single reaction vessel (one-pot method). According to the invention, the above is achieved, whereby the neutralisation of the alkaline lysate component is carried out in one and the same container by the addition of potassium phosphate and hence one component of the aqueous biphasic system is already present and the second component of the aqueous biphasic system is a PEG with a molecular weight of calculated average from about 600 g/mol to 1000 g/mol, however, said second component is preferably formed by a mixture of PEG 600 and PEG 1000.
