A method of screening a nucleic acid encoding a protein interacting with a target substance from a nucleic acid library which comprises: the step of producing a library of assigned molecules each having a protein and a nucleic acid encoding the protein which are ligated via a cleavable linker under such conditions as not changing the base sequence of the nucleic acid; the step of mixing the assigned molecule library with the target substance; the step of separating an assigned molecule bonded to the target substance; the step of cleaving the linker of the assigned molecule thus selected under such conditions as not changing the base sequence of the nucleic acid to thereby release the nucleic acid; and recovering the thus released nucleic acid. According to this method, an assigned molecule bonded specifically to a target substance can be screened at a high efficiency. |
