Disclosed is a method of manufacturing a controlled release formulation by use of a pelletizer, including the steps of extruding a drug-containing combination to a filament, and molding the filament to a pellet. The combination having suitable ductility is obtained by dissolving at least a drug and a hydrophilic polymer or a surfactant for use in assisting dissolution of the drug in a solvent, to form a mixture, and combining the mixture with an inert diluent and/or a dissolution inhibitor. The method is advantageous in that drug loss during production of the formulation is prevented, whereby the formulation having a uniform amount of the drug can be produced. |
