titleOfInvention Process for determining endotoxin level in hemoglobin solutions
abstract Procedure for determination of endotoxin level in the hemoglobin solution which includes application of LAL kinetic-turbidimetric method at the 650nm and 660nm wave length by using the corresponding filters and corresponding modifications of the 10%w/v hemoglobin sample. 10%w/v hemoglobin solution has been diluted to concentration of 10mg/ml and then incubated within the temperature range of 60-80 C/10-20 minutes. Incubated solution has been diluted to concentration of 2,5mg/ml and then tested by application of LAL kinetic-turbidimetric method at the 650nm and 660nm wave lengths together with application of corresponding filters. Endotoxin concentrations have been read from the standard curve which has been obtained for LAL kinetic-turbidimetric method at the 650nm and 660nm wave lengths. Test is valid if the value of the positive product control is within the range 50-200% in relation to the pure sample, and hemoglobin solution is pyrogen-free if the endotoxin level detected in the sample is 0,595EU/ml ( 5,95EU/g hemoglobin).
