titleOfInvention | Method for the determination of cytosine methylation in cpg islands |
abstract | A method for the determination of cytosine methylation in DNA samples is disclosed. The DNA is firstly extracted from the sample and bonded to a surface. In the second method step a genomic DNA sample is preferably treated with a bisulphite (= disulphite, hydrogen sulphite) in such a way that all non-methylated cytosine bases are converted to uracil, whilst the 5-methylcytosine bases remain unchanged. In the third method step one or several oligonucleotides are hybridised onto the treated DNA as primer. In the fourth method step the hybridised primer(s) are lengthened in a polymerase reaction. Marked guanine nucleotides are preferably used which are then essentially only incorporated when cytosine bases are still present in the treated DNA. Thereafter the measure of inclusion of guanine bases and thus the number of incorporated markings is proportional to the methylation in the sample under investigation. In the fifth method step the marked nucleotides not incorporated in the polymerase reaction are removed. In the sixth method step the number of markings in the fragment generated by the primary extension is approximately determined, whereby the signal intensity emitted by said markings is directly or indirectly measured. |
