abstract | A method for screening a compound for its ability to inhibit DNA adenine methylase (Dam) is disclosed. The method utilizes a double stranded DNA comprising at least one Dam target site. One end of the DNA is bound to a solid support such as the well of a microtitre plate. The other end is provided with a means for detecting whether the double stranded DNA comprising the Dam target site has been cleaved. In the assay, a compound to be tested is added to the well of the microtitre plate. Dam and a substrate therefor are added and if the compound is not an inhibitor of Dam the Dam will methylate the adenine residue of the Dam target site. By adding an enzyme that selectively cleaves the double stranded DNA at the Dam target site but which will not cleave a methylated Dam target site (or vice versa) it is possible to selectively cleave the detection means from the DNA. If the DNA is cleaved the detection means will be lost in a subsequent wash step whereas if the DNA is not cleaved the detection means will remain bound to the microtitre plate and can be used to generate, for example, a colour reaction which can in turn be used to determine whether the compound under test is an inhibitor of Dam. Also disclosed is a kit for conducting such an assay. |
