abstract |
A method for synthesizing peptides is disclosed wherein enzymes are used in both the coupling and deprotecting steps. The coupling takes place at a first temperature where the enzyme catalyzing deprotection is not active, and deprotection takes place at a second temperature where the enzyme catalyzing coupling is inactive (Figure 1). By cycling between the first and second temperatures control of coupling and deprotection can be obtained. Also disclosed is a screening method for identifying an enzyme suitable for deprotection in the peptide synthesis method. |
