A clone PLACE 6002312 having a structure characteristic to G protein-coupled receptors is found out from a human placenta-origin full-length cDNA library prepared by the oligo-cap method. As the results of data base search, PLACE 6002312 shows the highest homology with histamine H2 receptor. By analyzing the expression of the PLACE 6002312 gene in human tissues, it is found out that this gene is expressed in many tissues such as heart, liver and ovary. As the results of ligand-binding assay, it is found out that histamine is one of the ligands of PLACE 6002312. Further, a full-length cDNA of a mouse homolog of PLACE 6002312 is isolated and a structure characteristic to G protein-coupled receptors is found out in its anticipated amino acid sequence. It is expected that PLACE 6002312 might be applicable to screening a pharmaceutically useful agonist or antagonist, diagnosis of various diseases based on abnormalities in the activity or expression thereof, etc. |
