abstract | The present invention relates to a sustained release composition and a method of preparing and administering the sustained release composition. The method of the invention for preparing a composition for the sustained release of a biologically active agent, comprises the steps of: (a) forming a dispersed system comprising a biologically active agent, a water soluble polymer and a first solvent for the water soluble polymer; (b) atomizing the dispersed system to form droplets; (c) freezing the droplets to produce frozen droplets; (d) removing at least a substantial portion of the first solvent from the frozen droplets to produce a dried substance comprising the biologically active agent and the water soluble polymer; (e) preparing a mixture comprising the dried substance, a biocompatible polymer, and a second solvent for the biocompatible polymer and the water soluble polymer which is a non-solvent for the biologically active agent; (f) fragmenting the biologically active agent of the mixture; (g) removing the second solvent from the mixture thereby forming a solid biocompatible polymer matrix having dispersed therein the biologically active agent and the water soluble polymer. |
