abstract |
The present invention relates to expression modules that can be used to assemble integrating and autonomously replicating vectors for use with filamentous fungi. The vectors constructed with these modules are all based on the backbone of the E. Coli plasmid pUC18. They can therefore be used to perform most standard DNA manipulations in the same plasmid that is introduced into the fungus. This system of vectors can be used to study promoter sequences, express heterologous and homologous proteins, and identify vector and host strain mutations that alter protein expression. A subset of these vectors, which include the Aspergillus niger LacA coding region as a reporter gene, were used to develop a high-throughput indicator plate-based screen that could identify host and vector mutants that increase the expression of the secreted reporter protein, LacAp. Finally, the vectors and strains developed can be used to develop strains that can express the coding region for essentially any homologous or heterologous protein at high levels. |
