abstract | Disclosed are methods of introducing a mutation into a target nucleotide sequence. The method facilitates the introduction of more than one mutation sequentially into the target sequence using multiple rounds of mutagenesis, allowing the appropriate mutated sequences to be positively selected after each step. More than one target primer may be used simultaneously to introduce more than one mutation into the target sequence. Preferably hosts carrying the selection sequence and the mutated selection sequence can be simultaneously positively selected and distinguished on the basis of colour e.g. arising from mutations in the alpha-complement factor or an active variant thereof e.g. as encoded by lac Z. |
