A method of Agrobacterium-mediated genetic transformation using seedlings has been found which is applicable to dicots and monocots capable of being transformed by Agrobacterium. The transformation method utilizes vacuum-infiltration to introduce the Agrobacterium T-DNA carrying a gene of interest into the seedlings. Upon maturity, seeds collected from the infiltrated seedlings are germinated, and progeny carrying the transgene are selected. This transformation method produces progeny exhibiting stable inheritance of the transgene without the need for regeneration methods such as somatic embryogenesis or organogenesis. |
