abstract An object of the present invention is to provide a method for amplification of long nucleic acid, wherein the method allows nucleic acid fragments containing the same nucleotide sequence information to efficiently amplify at the same base length. The present invention relates to a method for amplification of long nucleic acid sequence, wherein the method uses primers being modified at the 5′ end with a phosphate group and performs a cooperative reaction using DNA polymerase and DNA ligase.
