Detection and/or quantification method of target molecules on a solid support |
abstract |
The invention relates a method for detecting and/or quantifying different target molecules being bound at different locations (spots) of a surface of an optically transparent solid support having a refractive index n 1 by specific interaction with capture molecules being immobilized on the solid support surface, said solid support being in contact with a solution having refractive index n 2 , whereby n 1 >n 2 and wherein the target molecules are in contact with the capture molecules, said method comprising the steps of:n a) illuminating homogeneously the surface of the support on which the target molecules are bound, thereby causing homogeneous excitation of the target molecule; b) detecting light emitted from the target molecules in at least four different locations (spots) in the following way: collecting the emitted light through a side of said support which is inclined relative to the surface of the support on which the target molecules are bound, focusing the emitted light by a lens on a detector surface which is positioned at an observation angle θ obin relative to the normal to the said solid support surface in the support, such that 90°>θ obin >sin −1 (n 2 /n 1 ), obtaining an image of at least 4 different locations (spots) being spatially discriminated onto the surface of the detector. |
