Provided are isolated antibodies or fragments thereof which bind a peptide consisting of residues His 73 -Val 74 -Leu 75 -Leu 76 -Thr 77 -His 78 -Thr 79 -Ile 80 -Ser 81 -Arg 82 -Ile 83 -Ala 84 -Val 85 -Ser 86 -Tyr 87 -Gln 88 -Thr 89 -Lys 90 -Val 91 -Asn 92 -Leu 93 -Leu 94 (peptide 309) of mature human TNF-α. The antibodies and fragments thereof may be used to identify antibodies capable of binding in the region of mature human TNF-α of amino acid resides 73-94. Antibodies or fragments thereof which bind particular regions of mature human TNF-α are shown to elicit particular biological activities dependent upon the particular region wherein binding occurs. |
