abstract | Methods for producing a population of distinct aRNA molecules from an initial population of distinct mRNA molecules are provided. In the subject methods, an initial mRNA sample is contacted with a population of distinct tagged antisense molecules to produce a population of hybrid or duplex molecules. The resultant population of hybrid molecules, or template derivatives thereof, is then contacted with a DNA dependent RNA polymerase to produce the population of distinct aRNA molecules. Also provided are kits for practicing these methods. The subject methods find use a variety of different applications in which the preparation of aRNA is desired, e.g., the preparation of nucleic acid targets for use in array based hybridization applications. |
