The invention relates to a method for producing recombinant human c-peptide, including cultivation of Escherichia coli producing strain, bacterial cell disintegration, inclusion bodies containing hybrid protein, dissolving them in urea and diuretic buffer, purification of the reconstituted hybrid protein, its cleavage by trypsin and carboxypeptidase B, the treatment of which is carried out simultaneously, while the products of trypsinolysis are separated by chromatography on SP-Sepharose balanced with 0.03-0.1 M ammonium-a etatnym buffer pH 3.6 containing 3M urea eluting with a linear gradient of potassium chloride from 0 to 0, 5 M in the starting buffer, followed by purification and yield the desired product. |
