abstract | (57) The invention relates to genetic engineering, in particular to the construction of recombinant plasmid DNA for the production of interferons. Plasmid p9A2 or E79E9 was digested with restriction enzyme Ava II, the resulting insert to DNA was digested with Sa and 3A, a 189 bp fragment was isolated, plasmid pER33 was digested with restriction enzymes EcoR I and Pvu II, a 389 bp fragment. then digested with enzymes 5a and 3A, a 108 bp fragment is isolated, which is ligated with a 189 bp fragment, the resulting DNA is treated with Hind III and ligated with Hind III with the linearized plasmid ER 103, the resulting recombinant DNA is transformed into an E. coli strain HB 101, the resulting plasmid pRHW 10 is digested with Bam H1, incubated in the presence of the Klenow fragment of DNA polymerase 1 and four deoxynucleoside triphosphates, the plasmid is treated with the Nco I enzyme, and a large Nco I - Alu I fragment of plasmid p9A2 or E79E9 is isolated, which is obtained as a result of processing the plasmid Ava II followed by hydrolysis with Nco I and Alu I, the recombinant plasmid DNA is transformed into E. coli HB 101.
