abstract | A method of modeling pneumopathy in systemic autoimmune disease is the introduction of a complete Freund's adjuvant with a solution of spleen deoxyribonucleic acid in cattle. Nonlinear white rats with an average weight of about 200-250 g under ether inhalation anesthesia are injected into the root of the tail with Freund's adjuvant and a solution of splenic DNA of cattle (5 mg / kg body weight). Sodium azide (2 mg / kg) and sodium deoxyribonucleinate (20 mg / kg) are administered intraperitoneally over three days. One week after the start of the experiment, cyclophosphamide (100 mg / kg) is additionally injected intramuscularly, and a complete Freund's adjuvant and a solution of spleen DNA of cattle (2.5 mg / kg) are added to the root of the tail. From the following day, sodium deoxyribonucleinate (20 mg / kg) and cyclophosphamide (100 mg / kg) were administered intramuscularly daily to the animals for one week. Throughout the experiment (14 days) rats are in a chamber where the air contains 1% ammonia solution (0.6 l / m3), 50% benzene solution in gasoline (1.3 l / m3), 100% formalin solution (2, 5 l / m3) and 20% solution of 2-mercaptoethanol (0.1 l / m3). After 2 weeks from the start of the experiment on the background of intra-abdominal nembutal anesthesia (50 mg / kg) by decapitation, the animals are removed from the experiment. The lungs are fixed in 96 ° ethyl alcohol. Sections are stained with hematoxylin-eosin and Van Gizon, and morphological examination of alveoli, bronchi and vessels, in the presence of changes in which the method is considered reproducible. |
