abstract | A method of obtaining purified and soluble recombinant human interleukin-10 synthesized by E. coli BL21 / hIL10 producing strain includes bacterial cell deposition upon completion of biosynthesis by centrifugation and destruction of bacterial cells and chromosomal DNA by enzymatic attack in the presence of lysozyme. The inclusion bodies of the inclusion bodies were centrifuged and sequentially washed the inclusion bodies with solutions containing non-ionic detergents and urea-containing solutions by suspending the sediment with subsequent precipitation by centrifugation, solubilizing the inclusion bodies in the presence of guinea. The hIL-10 is renatured by replacing the buffer by column gel gel filtration chromatography in the presence of L-arginine hydrochloride and non-ionic detergent, followed by purification of the protein under native conditions by anion exchange and affinity chromatography. |
