abstract | A serum-free culture of autologous mesenchymal stem cells in vitro includes following steps: collecting blood; mixing blood with a reaction reagent including an anticoagulant and a thixotropic colloid; centrifuging the blood including the reaction reagent to make the blood to react with the reaction reagent to form into three layers; taking an upper layer which is a platelet plasma layer and mixing it with the anticoagulant to obtain an autologous growth factor; adding the autologous growth factor to a cell culture at a volume ratio of 0.25 to 2%; providing autologous mesenchymal stem cells; and culturing the autologous mesenchymal stem cells in the cell culture supplemented with the nautologous growth factor. The culture can replace the traditional heterologous serum culture, and maintain the characteristics of the autologous mesenchymal stem cells. |
