abstract The present invention provides a chimeric protein comprising a factor VIII (FVIII) polypeptide and an XTEN polypeptide. The FVIII polypeptide may have a smaller affinity for Von Willich's factor (VWF). An XTEN polypeptide of an appropriate size is inserted into a specific position in the FVIII polypeptide to extend the half-life of the FVIII polypeptide. The invention also includes nucleotides, vectors, host cells, and methods of using such chimeric proteins, for example, to treat bleeding diseases and disorders.
