abstract |
The present invention provides a novel yeast mutant strain which can be utilized to produce a glycoprotein by adding a sugar chains with the same structure as the sugar chains produced by mammalian cells to the asparagines residue of protein, and a method for preparing the sugar chains and the glycoprotein by using said mutant strains with glycotechnological approach. The novel bred auxotrophic triple mutant strain or auxotrophic quadruple mutant strain according to the present invention, it is possible to produce a neutral sugar chain the same as the high-mannose type sugar chains which produced by the mammalian cells like human being, or a glycoprotein containing said neutral sugar chains in quantity and higher purity. Also, it is possible to produce a mammalian-typed sugar chain of high-mannose type, hybrid type or complex type, or a glycoprotein containing said sugar chains efficiently by transducing the mutant strains into a biosynthetic gene of mammalian-typed sugar chains. |
