abstract | The present invention relates to the use of biodegradable microspheres which release a radio-sensitizing anticancer agent, for the manufacture of a medicinal product intended to be used simultaneously or separately in time or spread out over time with a radiotherapy, for the treatment of glioblastoma. The use of the biodegradable microspheres according to the invention achieves a patient survival time of at least 90 weeks, while maintaining a therapeutically effective concentration over time in the parenchymal space. The microspheres used preferably contain 5-fluorouracil coated with poly(d,1-lactic acid-co-glycolic acid). The microspheres are implanted by intratissue injection, after excision of the tumor, into the walls of the operating locus. Radiotherapy focused on the tumor volume is dosed at 60 Gy over about six weeks. The present invention also relates to a process for preparing biodegradable microspheres by emulsification-extraction, as well as to a suspension containing biodegradable microspheres which may be obtained by this process. |
