abstract A stable kit formulation has been developed for preparation of 99mTc-TRODAT-1 with high yield. New formulation comprises HC1 salt of TRODAT-1 to improve the stability and solubility of free TRODAT-1. Mannitol is used to stabilize the lyophilized component. Following reconstitution with 99mTc-pertechnetate and heating, 99mTc-TRODAT-1 can be obtained in a neutral solution with greater than 90% radiochemical purity determined in 1.5 hours. The shelf life of the lyophilized kit lasts at least 2 months when it was stored at 33 DEG C. Brain SPECT study on primates verifies the clinical potential of 99mTc-TRODAT-1 preparation with this novel kit formulation.
