abstract | Claims of this patent are (1) production of anti-humanLT β Rmonoclonal antibodies(mAb). Clone 25A8 is agonistic, while clone 1B8 is antagonistic mAb; (2) production of LTβR.Fc fusion gene by the ligation of extracellular domain of human LTβR with the portion of human IgG1, then expressed either by baculovirus system or CHO cells; (3) using the LTβR.Fc fusion protein as antigen to obtain monoclonal antibodies against LTβR. In conjunction with interferon-gamma, clone 25A8 could induce apoptosis (or programmed cell death) in several tumor cell lines, therefore it could be used for anti-cancer therapy. The LTβR.Fc fusion protein could be used to purify soluble form lymphotoxin Α1β2 or Α1β1 heterotrimer, or lymphotoxin Α1β2, Α1β1 or lymphotoxin γ/LIGHT in vivo. Antibody generated against the recombinant LTβR.Fc fusion protein produced by baculovirus system can recognize the three-dimension epitope of human LTβR. This will ensure the binding specificity to antibody to the native form of human LTβR. The approach could be applied to other receptors whose extracellular domain is located on the N-terminal region. |
