abstract | The invention relates to a protodrug made of L-dopa in coupled with D-phenylglycine or D-p-hydroxyphenylglycine and the dipeptides thereof that is capable of enhancing the oral bioavailablity of the protodrug and delaying the internal metabolic rate of decarboxylation for the L-dopa to prolong its half-life. In the experimental animal model of anti-Parkinson's disease, compound 2d can mitigate the rotation behavior of the rat with damaged gray matter of the brain that is stimulated by (+)toluene-iso-propylamine. The compound 2d has an anti-Parkinson's disease activity equivalent to L-dopa and does not cause the contraction of the pulmonary artery and vasa deferens, indicating that no dopamine-like side effect such as hypertension leading cardiovascular diseases. Such drug is useful in the treatment of patients suffered from Parkinson's disease with hypertension. |
