Method for enhancing the transcriptional regulation ability of SUPT4H in a nucleotide repeat sequence of a cell |
abstract |
The invention provides a method for regulating a gene containing a nucleotide repeat sequence expansion cell in a cell, which comprises: inhibiting the biological activity of SPT4 or SUPT4H; and regulating the formation of R-loops. The inhibition step can effectively reduce the expression of the nucleotide repeat expansion gene and the regulation step can further enhance the effect of the inhibition step. The inhibition step and the regulation step interfere with the ability of RNA polymerase Ⅱ to transcribe the DNA template through the R-loops generated by the nucleotide repeat sequence, thereby achieving the purpose of regulating gene expression. |
