abstract |
This article reports a method for generating multispecific antibodies by a half-antibody exchange reaction. The half-antibody exchange reaction is performed between two 2 / 3-IgGs, and half of the two 2 / 3-IgGs are asymmetric Perturbation mutations and lose stability, thereby promoting the production of full-length bispecific antibodies that are correctly assembled. This method can be performed in the absence of a reducing agent and does not require the presence of hinge disulfide bonds in the original 2 / 3-IgG. |
