abstract |
The present invention provides a multi-expression target protein system comprising dihydrofolate reductase-deficient CHO cells, a folic acid antagonist, a target protein expression vector and a CRISPRi expression vector, wherein the target protein expression vector comprises a target protein exhibiting a cassette and a DHFR expression cassette. The CRISPRi expression vector contains gRNA cassettes and dCas9 expression cassettes. The large amount of the expressed target protein system can greatly increase the yield of the target protein and shorten the time for gene amplification. |
