abstract A monoclonal antibody against PD-1 and a method for obtaining the same, comprising the amino acid sequence encoding the variable region and the complementarity determining region of the antibody, the obtaining method and the use of the monoclonal antibody. The present invention selects a monoclonal antibody against PD-1 from a fully synthetic antibody library, and then performs affinity maturation by a phage library chain replacement method, and firstly mutates the CDR1, 2, and 3 regions of the light chain of the monoclonal antibody obtained by the primary screening. After the library was screened, the monoclonal antibody with higher affinity was selected, and the CDR1, 2, and 3 regions of the heavy chain were screened, and the high-affinity monoclonal antibody against PD-1 was finally screened. The PD-1 fully human antibody obtained by the present invention has good affinity for PD-1 and can inhibit the binding of PD-1 to its ligand, and can be used for the treatment of tumor, inflammation and autoimmune diseases.
