abstract | This disclosure discloses methods for preparing modified functional complexes. First, a biotinylated peptide with a molar ratio of 1:4 to 16:1 is slowly added to the avidin-like particles to form a plurality of avidin-biotin with different valences. A peptide complex wherein each biotinylated peptide comprises a set of amino acid tag sequences. Thereafter, the avidin-biotin-peptide complex is eluted with a nickel affinity column and an imidazole concentration gradient to obtain a first single valency avidin class - biotin - which is mainly composed of the first valence A first eluate composed of a peptide complex. Next, the first single valency avidin-biotin-peptide complex is mixed with the functional particles under appropriate conditions to form a first single valence biotin with a first valence-binding peptide Modified functional particles. Also disclosed herein are methods for preparing a single valence modified avidin-like particle. |
