abstract |
High resolution A protein chromatography using chaotropic agents and pH gradients or pH-level elution buffers results in improved peak resolution between closely related molecular species. Bispecific antibody containing A protein-binding-ablative substituted CH3 region paired with A protein-binding CH3 region and A protein-binding with A protein-binding-ablative-substituted CH3 region were characterized by high peak resolution - a single specific antibody that ablates the substituted CH3 region and a single specific antibody that contains the A protein-binding CH3 region paired with the A protein-binding CH3 region. Useful chaotropic agents include magnesium chloride and calcium chloride. |
