abstract |
The present invention relates to a method of mimicking a germinal center response to generate antibodies having different binding properties or higher affinities directly in a fusion tumor. To facilitate the rapid affinity maturation of antibodies, a controllable activation-induced cytidine deaminase (AID) expression system was developed which induces somatic high frequency mutations in fusion tumors. By affinity-activated cell sorting techniques, high-affinity antibodies can be selected from fusion tumors that preferentially bind to the fluorescently labeled antigen. The present invention also relates to a novel method of producing a fusion tumor having a tunable antibody affinity by generating a myeloma fusion partner having a controllable AID expression. |
