abstract |
The present invention discloses novel antibodies and fragments thereof that specifically bind to CD4. The present invention discloses anti-CD4 antibodies having an amino acid substitution in the hinge region that prevents the formation of intrachain disulfide bonds, resulting in antibody molecules with surprisingly improved bivalent stability. Increased stability facilitates manufacturing and provides increased in vivo stability. The present invention also discloses antibodies having an amino acid substitution that reduces binding to Fc receptors and ADCC activity and also produces surprising effects on CD4 internalization. In addition, the present invention discloses genes for such antibodies that have been modified to have an increased performance over their unmodified counterparts. |
