The method comprises the steps of: providing a single-stranded circular DNA primer with a predetermined length, and amplifying a single-stranded circular DNA primer into a single-stranded DNA nanotemplate by a constant temperature nucleic acid amplification reaction, and fixing one end of the single-stranded DNA nanotemplate to the surface of the substrate; And adding a single-strand DNA probe, one end of each single-stranded DNA probe is bound to a nanoparticle, and a single-stranded DNA probe is bonded to a corresponding sequence on a single-stranded DNA nanotemplate to form a nanoparticle chain. The method for producing a nanoparticle chain of the present invention further comprises providing a fluid, and the flow of the fluid controls the direction in which the nanoparticle chains are arranged. Among them, the nanoparticle spacing on the nanoparticle chain can be adjusted by the reaction temperature or a single-strand DNA probe to which unbound nanoparticles are added.
