abstract The invention discloses a single-step purification method for recombinant Helicobacter pylori neutrophil activating protein (HP-NAP), the method comprising the steps of: providing a sample to be purified containing HP-NAP, wherein the sample to be purified is taken from a sample a protein expression system capable of expressing recombinant HP-NAP; the sample to be purified containing recombinant HP-NAP is purified by a diethylaminoethyl (DEAE) ion exchange resin at a predetermined pH interval; and collected via the above And a flow through fraction and a wash fraction, wherein the effluent and the cleaning solution comprise recombinant HP-NAP of a predetermined purity. The present invention utilizes a high affinity DEAE Sephadex ion exchange resin to purify the recombinant HP-NAP of the water-soluble protein in a single step, thereby purifying the recombinant HP-NAP with low endotoxin residue from the Bacillus subtilis expression system, and purifying it. The recombinant HP-NAP has a typical alpha-helix secondary structure and activates neutrophils to produce reactive oxygen species.
