abstract The invention discloses a human second type topoisomerase (hTOPII)-DNA binary complex, a preparation method thereof and application thereof. The hTOPII-DNA binary complex comprises a hTOPII site containing a hTOPII core region (hTOPIIcore) and a synthetic double-stranded DNA complexed with a hTOPII site. The synthetic double-stranded DNA has a first strand of DNA comprising a 5'-NNNCCGAGCNNNNGCTCGGNNN-3' sequence (SEQ ID. NO.: 1) at nucleotide positions 3 to 20, wherein N is adenine, thymine, cytosine or Guanine, and the second strand of DNA complementary to the first strand of DNA.
