abstract | Disclosed is a method for recombinant botulinum neurotoxin A vaccine production by baculovirus expression system, comprising the steps of: (a) providing a first vector and a second vector and a reporter protein; (b) cotransfecing the first vector and second vector to a cell for recombination; (c) screening the recombinant baculovirus by the reporter protein; and (d) infecting insect cells with the recombinant baculovirus and producing BoNT/A vaccine. The recombinant BoNT/A HC produced by the present invention has higher solubility. The discosed expression system has a higher yield without changing the gene codon and would not produce inclusion body and endotoxin. In the future, the expression system has a great potential to be a novel recombinant botulinum neurotoxin A vaccine production plateform. |
