abstract The present invention relates to insecticidal gene and protein from pseudomonas sp. TKU015, production method and application thereof. The production method includes the following steps: (1) culturing Pseudomonas sp. TKU015 under a condition that is optimally suitable for nattokinase production; (2) recovering the culture supernatant to isolate nattokinase solution; (3) performing molecular biological experiments on the amino acid sequence fragments obtained by adopting liquid chromatography-tandem mass spectroscopy(LC-MS/MS) analysis on the nattokinase solution to find out related genes; (4) designing suitable primers to find out insecticidal protein gene sequence fragments in pseudomonas sp. TKU015 by polymerase chain reaction(PCR); (5) using molecular biological techniques to further obtain a full-length gene of insecticidal protein; (6) transferring the insecticidal protein gene to Escherichia Coli host for over-expression; and (7) using a purification system on vectors to isolate and purify insecticidal protein for obtaining a purified insecticidal protein. The present invention also provides insecticidal application of insecticidal protein from pseudomonas sp. TKU015.
