titleOfInvention | Production method of peptide thioesters |
abstract | The issue of this invention is to provide a novel method for converting a peptide chain into a peptide thioester through a chemical manner. The inventors of this invention focus on the cysteine residue in the peptide chain and then found that: a –C(=X)-R.sub.1 group is introduced into the thiol group of the cysteine residue. A compound reaction having the leaving group represented by –NH-C(=Y)NHR.sub.3 in the organic solvent is performed. Accordingly, the –NH-C(=Y)NHR.sub.3 group is added on the carboxyl group of the N-end peptide bond of the cysteine residue. As a result, the peptide bond is cleaved, and the peptide fragment at the C-end is removed. The invention further found that: with the following so-called thiol exchange reaction: since the thiol is reacted with a peptide chain added with the –NH-C(=Y)NHR.sub.3 group in a buffer solution, the carbonyl carbon bonded with the –NH-C(=Y)NHR.sub.3 group is now bonded with the thiol group having the thiol, and the –NH-C(=Y)NHR.sub.3 group comes off, and the peptide chain is converted into a peptide thioester. |
