abstract This invention is the first time to separate, purify nucleic acid molecules with clinical significance from the human stool. The nucleic acid molecules purified by the method can be applied to detect the situation of measuring the invading cancer patient of carcinoma of the rectum of large intestine. Its key steps include the following methods: First Utilize the Guanidinium Thiocyanate Buffer to liquidize and unfreeze the fast refrigerated stool and disperse in a polypropylene test tube. Second, cultivate, ultracentrifuge and extract the supernatant and purify with pure alcohol and column. Then fetch 1 to 2 micrograms RNA and 0.5 micrograms of oligo-dT to carry on RT-PCR. Third, amplify CK18 with RT-PCR. Evaluate the stool CK19 mRNA of patient with CRC and healthy person as contrast. And the mRNA got by three above-mentioned steps, has steady degree that can be up to 24 hours and enable examining the body to transport unlikely total RNA got by previous other methods likely to degrade. Besides it can also increase the credibility of the experiment , and can improve the uniqueness of the human gene displays . We detect the steady behavior of cell's keratin gene cytokeratin No. 18, one of housekeeping gene, while examining the human stool. And only the cell's keratin gene No. 19 < cytokeratin 19> at the stool of large intestine carcinoma patient has the high expression. But it does not have this phenomenon in the stool of normal person being examined. In view of the above, the purified stool nucleic acid molecules can carry on various kinds of differences and detecting and examination of the gene. And this technology can further make the nucleic acid molecule with clinical significance existed in the stool apply to screen the person who didn't get invading large intestine carcinoma of the rectum.
