abstract The invention relates to urea derivatives, in particular the preparation of a mixture of 9- (2-chloroethyl) -7- (or 9) nitroso-L-homocytrulline isomers (I and II) used in pharmaceuticals. The goal is to increase the yield of the target product with a certain isomeric composition. Synthesis is carried out by treating lysine KOH and copper sulfate in an aqueous medium, followed by the introduction of the carbamoylating agent 1,3-bis- (2-chloroethyl) -1-nitrosourea at 32-36 ° C, decomposition of the complex with oxalic acid in the presence of HCl, nitrosation of NaNO into HCl at pH 1 - 2. The resulting mixture of isomers is divided in two stages: from a water-alcohol solution of hydrochloric acid, followed by neutralization with aqueous NH4 to pH 5.2 - 5.4 at minus 2 - plus 2C; crystallization at minus 10 - minus 12C and a mass ratio of the copper complex, water and alcohol in the first stage 1: 4.5 - 5: 2 - 2.4 and in the second - a mixture of isomers, water and alcohol 1: 3.8 - 4, 2: 1,1 - 1,3. In this case, the average yield of a mixture of isomers with a ratio of compounds I and II of 23–27 (%): 47–73 (%), the content of the basic substance 98–99.7% is 38.9%.
