abstract | Usage: Mus musculus L. BCKK (P) No. 420 hybrid cultured cell strain producing monoclonal antibodies (MCA) to common antigenic determinants of human and porcine insulin, used for quantitative determination of insulin using immunoassay (ELISA) or immunoblotting . Summary of the Invention: Obtaining an ICA with an increased insulin affinity constant. The strain is obtained by hybridization of splenocytes of Balb / c mice with Sp2 / 0 myeloma cells. Culture media: D MEM with 10% SEC. Passage scheme: once in 4 to 5 days in vitro. In the form of ascites, the cells are intertwined for 8-10 passages. The concentration of ICA in the culture fluid on the 7th day of cultivation was 40 µg / ml, and in ascites - 5-8 mg / ml. Using ELISA using the obtained mAb, 30 pg hormone / ml are detected. |
