titleOfInvention Strain of hybridous cultured mammalian cells mus musculus l - a producer of monoclonal antibody to pyrophosphatase of escherichia coli
abstract The invention relates to biotechnology and can be used to create an enzyme immunoassay based on the pyrophosphatase-anti-pyrophosphatase complex. The aim of the invention is to obtain a hybrid strain of cultured mouse cells producing monoclonal antibodies (MAb) to Eseherihia colt pyrophosphatase. The strain is obtained by hybridization of splenocytes of B1L / c mice immunized with the E. coli pyrophos phatase preparation. with myeloma cells. P3x63-Ad8.653 mice. The cells are cultured in Schom medium (Dulbecco's modified Iskovo medium) with the addition of 10% fetal bovine serum at 37 ° C in an atmosphere of 5% COa. The strain is designated PFB1 and deposited under the number of CASA (P) 4220. The strain produces a monoclonal IgCI antibody of the isotype, which binds to E. pF phosphorase. The content of MCA'V in 1 ml of culture medium and 1 ml of ascites liquid is, respectively, 11-12 µg and 4-5 mg. MCA can be used to create an enzyme immunoassay based on the pyrophosphatase-anti-pyrophosphate complex. 2 tab. •
