abstract The invention relates to hybridoma technology and virology and can be used to improve the quality of the secreted eukaryotic RNA polymerase enzyme. The aim of the invention is to obtain a hybridoma strain producing monoclonal antibodies (Mon AT) to bacteriophage T7 RNA polymerase. The strain is obtained by hybridization of silocytes of BALB / C mice immunized with the purified preparation of RNA polymerase, bacteriophage T7, with cells of the mouse myeloma of the X 63 line. AG 8.653. Cells were cultured in DMEM medium with fetal bovine serum (10%), glutamine (2 mmol), sodium pyruvate (1 mmol), 2-mercaptoethanol (0.05 mmol), penicillin / streptomycin (1000 units / ml). The frequency of passaging 2-3 times a week, sowing - a dose of 150 thousand cells per 1 ml, the multiplicity of sowing 5-10 times. The productivity of strain 10 (by ELISA) in culture medium and 3 x 10 in ascitic fluid. Mon AT, secreted by the strain, specifically interacts with the bacteriophage T7 RNA polymerase.
