titleOfInvention Strain of hybrid cultivable mus musculus l cellis used for producing monoclonal antibodies to human collagen
abstract This invention relates to hybridoma technology and can be used in medicine and biology. The aim of the invention is to obtain a hybrid strain of cultured animal cells MUS MUSCULUS L producing monoclonal antibodies (MON AT) to human collagen type I and III. The strain is obtained by hybridization of spleen cells of a BALB / C mouse, immunized with human type I collagen, with X63 P3 / Ad 8.653 mouse myeloma cells. The strain was deposited as VSKK number (P) N163 D. The hybridoma was cultivated in the DMEM medium with bicarbonate and 20% fetal bovine serum, 2 mM L-glutamine, 2 mM pyruvate sodium, 100 µg / ml kanamycin, 5 µg / ml Fungison, 0 , 05 mM 2-mercaptoethanol at 37 ° C in an atmosphere of 5% CO 2 . The strain produces MONs of the AT class YGM, which specifically bind to human collagens type I and III. For 3-4 days of cultivation, the concentration of MOH of AT is 1-10 µg / ml in culture medium and 10-30 mg / ml in ascites fluid.
